- The effects of chronic administration of LH-RH agonists and antagonists on the menstrual cycle and endometrium of the rhesus monkey. Balmaceda, J. P.; Borghi, M. R.; Burgos, L.; Pauerstein, C. J.; Schally, A. V.; Asch, R. H. (Dep. Obstetr. Gynecol., Univ. Texas, San Antonio, TX 78284, USA). Contraception, 29(1), 83-90 (English) 1984. CODEN: CCPTAY. ISSN: 0010-7824. DOCUMENT TYPE: Journal CA Section: 2 (Mammalian Hormones) Regularly cycling rhesus monkeys (Macaca mulatta) were treated with: vehicle (Group 1); (6-D-tryptophan)-LH-RH [57773-63-4] (20 mg/day for 90 days, s.c.) (Group 2); and the LH-RH antagonist [N-Ac-D-Trp1,3, D-p-Cl-Phe2, D-Arg6, D-Ala10]-LH-RH [86044-76-0] (200 mg/day for 90 days, s.c.) (Groups 3). FSH [9002-68-0], LH [9002-67-9], estradiol (E2) [50-28-2], and progesterone (P) [57-83-0] were measured in the serum every 2nd day until 30 days past the discontinuation of drug administration. Endometrial biopsies were obtained on days 10, 40, 90, and 120 and processed for histol. exam and detn. of estrogen (E) and progesterone receptors. Animals of Group 1 presented regular cycles, whereas those in Groups 2 and 3 remained anovulatory throughout the treatment. Animals of Group 2 presented different degrees of endometrial hyperplasia during treatment, and animals of Group 3 showed either resting or atrophic endometrium. Administration of the LH-RH agonist increased E and P endometrial receptors, and the antagonist decreased P receptors. In both instances, reversal of the effects on the menstrual cycle and in the endometrium was obsd. 30 days after discontinuation of drug administration.
