Gastric adaptive relaxation is involved in the reservoir function of the proximal stomach. Ecabet sodium, synthesized from abietic acid, is widely used in Japan as an antiulcer drug. The purpose of this study was to evaluate the effects of ecabet on gastric adaptive relaxation in isolated guinea-pig stomachs. Changes in intragastric vol. and pressure were recorded in the presence of atropine and guanethidine. Gastric adaptive relaxation was triggered by luminal distention. Ecabet at the dose of 20 mmol/l in solns. at two pHs (pH 3.0 and 7.4) significantly lowered the threshold pressure needed for such relaxation and increased intragastric vol. 86408-72-2 is just another one chemical used in this study., but lower doses had little or no effect. The results suggested that this antiulcer drug might improve symptoms in patients with dysmotility-like functional dyspepsia. .

DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The distribution of TA-2711 in stomach was studied after oral administration of 14C-TA-2711 (100 mg/kg) to rats. At 6 h after dosing, most of the radioactivity was found in the lower intestine, cecum and large intestine. However, the radioactivity in the stomach was higher than that in the upper and middle small intestine. The concn. of the radioactivity in the glandular stomach was about 1.5 mg equiv TA-2711/g at 30 min after administration. Thereafter, it decreased gradually to about 60 mg equiv TA-2711/g at 6 h after dosing. In the radioluminograms of 6 and 24 h after dosing, most of the radioactivity was obsd. on the surface of gastric mucosa. After administration to rats with gastric ulcer induced by acetic acid, higher radioactivity was obsd. in the ulcerated tissues of the stomach than in the nonulcerated control tissues. In the stomach damaged by ethanol, the concns. of radioactivity in lesion parts were also higher than those in non-lesions parts.Except for chemicals metioned above, 86408-72-2 is also used. In the microautoradiograms of gastric mucosa damaged by ethanol, the developed silver grains were densely distributed in necrotic tissues. .
