DOCUMENT TYPE: Journal CA Section: 62 (Essential Oils and Cosmetics) Section cross-reference(s): 41, 63 Anal. of com. D & C Red No. 9 [5160-02-1] revealed the presence of 11 arom. azo compds. (subsidiary colors) including SudanI (1-phenylazo-2-naphthol) [842-07-9], a strong sensitizer. In the modified guinea pig maximization test, each of the subsidiary colors was a contact sensitizer, whereas D & C Red No. 9 itself was not. Arom. amine impurities in Red Lake C Amine (2-amino-4-methyl-5-chlorobenzenesulfonic acid) [88-53-9] were the source of formation of the subsidiary colors in D & C Red No. 9. D & C Red No. 9 synthesized with Red Lake C Amine from which these arom. amines had been removed as completely as possible, contained no subsidiary colors. Based on these results, a practical manufg. process for D & C Red No. 9 free of contact sensitizers was developed, based on treating the Red Lake C Amine with activated C in aq. NaOH soln. before diazotization. .

These impurities were isolated by high-performance liq. chromatog. and their structural formulas were confirmed by various anal. methods. Many kinds of arom. azo compds. (subsidiary colors) were found. The subsidiary colors were 11 kinds including Sudan 1. These subsidiary colors and the related compds. were synthesized for use as the std. samples. The subsidiary colors in Lake Red CBA showed pos. results with the modified guinea pig maximization test indicating that each of them was a contact sensitizer, while Lake Red CBA itself was not. Lake Red CBA synthesized with C acid [88-53-9] from which arom. amines were removed contained no subsidiary color. When Lake Red CBA was synthesized with the purified C acid to which 3 types of the typical arom. amines were added in fixed quantity, the corresponding arom. azo compds. were produced in proportion to the added amts. of arom. amines. The prepn. of Sudan III, Orange II, Quinoline Yellow WS, Quinizarin Green SS, etc., free from contact sensitive impurities, for use in cosmetic products is discussed.
