The effect was abolished by the benzodiazepine oxazepam [604-75-1], indicating that the behavioral effects were mediated by binding of the inosines to benzodiazepine receptors. In control calvaria the energy charge remained ~0.7 for 24 h. However, bones cultured with PTH or PGE2 showed a transient fall in the energy charge down to 0.5. This was not assocd. 58-64-0 and 73-24-5 are cas registry numbers of chemicals which are used as reagents here. with a fall in total adenine nucleotides. The rate of adenosine [58-61-7] metab. in cultured bone in vitro was studied by detg. the contents of adenosine, inosine [58-63-9], 5-deoxyadenosine [58-64-0], 2-deoxyinosine [890-38-0], and hypoxanthine [68-94-0] in the culture medium. There was a continuous increase in adenosine, inosine, and hypoxanthine as well as a disappearance of medium 2-deoxyadenosine that was accounted for by appearance of 2-deoxyinosine. The deaminating activity could be accounted for only partly by activity in the medium and thus the main activity probably resides in the bone cells. PTH (10-8M) did not alter the rate of disappearance of 2-deoxyadenosine or adenosine deaminease activity detd. in bone exts. Thus, 2 substances that increase Ca2+ mobilization from bone alter ATP utilization and(or) synthesis without significantly influencing adenosine prodn. or metab. .
