New neurotoxin from the venom of the large hornet (Vespa orientalis).Some chemicals with cas registry numbers like 89699-91-2 are also used. Tuichibaev, M. U.; Tashmukhamedov, B. A.; Magazanik, L. G. DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) A neurotoxin, designated as orientotoxin [89699-91-2], was isolated from V. orientalis by initial gel filtration of the venom on Sephadex G75. A highly toxic fraction III, which had a phospholipid-cleaving property, was further fractionated on cellulose column (CM-52) and then rechromatographed 2-3 times to isolate individual proteins. Orientotoxin was isolated from the 2nd-fraction (cellulose chromatog.), which had no phospholipid-cleaving property and the LD50 was 0.50 mg/kg. Fraction III, which had phospholipase activity, had an LD50 of 0.65 mg/kg. Orientotoxin, after further purifn., cleaved only 1-acyllysophospholipids. The max. enzymic activity of orientotoxin was obsd. at pH 7.5 in the presence of 1 mM Cu2+. Na deoxycholate and Triton X 100 totally inhibited the toxin activity. The toxin substantially inhibited nerve endplate potential. Orientotoxin is a presynaptic neurotoxin. .
