CAS No.89943-82-8 Cicletanine

Cicletanine is a novel antihypertensive agent that has been shown to possess vasorelaxant, natriuretic, and diuretic properties in preclinical and clinical studies. The mechanism(s) by which cicletanine induces these biological effects has not been definitely established.

Stimulation of potassium fluxes by natriuretic furopyridines in human erythrocytes. Braquet, Pierre; Garay, Ricardo (I.H.B., Le Plessis-Robinson, Fr.). Diuretics: Chem., Pharmacol., Clin. Appl., Proc. Int. Conf. Diuretics, 1st, 529-31. Edited by: Puschett, Jules B.; Greenberg, Arthur. Elsevier: New York, N. Y. (English) 1984. CODEN: 53NLAE. DOCUMENT TYPE: Conference CA Section: 1 (Pharmacology) Two different classes of diuretic drugs, (aryloxy)acetic acid (ethacrynic acid [58-54-8], tienilic acid [40180-04-9], and (-)indacrinone [56049-89-9]) natriuretic furopyridines (cicletanide [89943-82-8] and BN 50157 [91868-80-3]) stimulate K+ movements across human red cell membranes. The kinetic properties of this effect and strongly suggest that the effect is mediated by a K+ transport system with a specific binding site for some diuretic drugs. The stimulated K+ fluxes are resistant to ouabain, bumetanide and quinine, thus suggesting that they are not mediated by the Na,K-pump, Na+-K+-cotransport system or by the Ca2+-dependent K+-permeability. The intensity of stimulation of K+ fluxes is pos. correlated with the magnitude of the natriuretic effect.

The effect of cycletanide, a diuretic, on the platelet vessel wall interaction; its involvement in the arachidonic acid cascade. Bourgain, R. H.; Deby, C.; Andries, R.; Garay, R.; Braquet, P. (Lab. Fysiol. Fysiopathol., Vrije Univ. Brussel, Brussels 1090, Belg.). Biochem. Pharmacol., 33(23), 3917-18 (English) 1984. CODEN: BCPCA6. ISSN: 0006-2952. DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) In rats, topical superfusion with cycletanide (I) [89943-82-8] (10-3M) did not affect thrombogenesis-induced by topical superfusion with ADP [58-64-0] (4 ′ 10-4M) and arachidonic acid [506-32-1] (10-4M). However, I antagonized the enhancing effect of superfused tranylcypromine [155-09-9] (2 ′ 10-3M) on ADP and arachidonic acid induced thrombogenesis. Possible mechanisms involved in the actions of I on thrombogenesis are discussed.
