DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The acute effects of chymopapain (I) [9001-09-6] injection on the load-bearing characteristics of the facet joints of three L2-3 and three L4-5 level motion segments were studied in vitro. Immediately following I injection into the nucleus pulposus of the lumbar discs, a sepn. at the facet joints was noted, assocd. 9001-09-6 is also in the experiment. with an increase in disk height. After 3 compressive loading (20-140 kg)-unloading cycles, however, the disk height approached its preinjection level. After injection, all discs exhibited gross morphol. changes of the nucleus pulposus, although a significant portion of the nuclear material remained. These results are discussed with respect to the use of I in the treatment of sciatica and low back pain. .

DOCUMENT TYPE: Journal CA Section: 7 (Enzymes) Chymopapain (EC 3.4.22.6) possesses an essential and a nonessential thiol group, but enzyme prepns. produced hitherto always contained <2 thiol groups. 9001-09-6 and 2127-03-9 which are cas registry numbers are also used here. Starting from com. chymopapain or from papaya latex, pure enzyme having 2 thiol groups was prepd. The purifn. procedure included covalent chromatog. on activated thiol-Sepharose, which specifically binds thiol-contg. proteins. Elution was effected with cysteine in a stepwise manner, 1st at pH 5 then at pH 8. The pure enzyme had a mol. mass of 24,700 as estd. by SDS-polyacrylamide gel electrophoresis. Titrn. of the pure enzyme with 2,2'-dipyridyl disulfide at pH 9 exhibited a biphasic curve. This indicated that under the conditions employed the nonessential thiol group is more reactive than the essential one, which is a characteristic feature of chymopapain B. On the other hand, the magnitude of the rate const. of the same reaction at pH 4, as well as its pH dependence, was characteristic of chymopapain A. The enzyme with the hybrid kinetic properties was denoted as chymopapain S. Apparently, various forms of chymopapain can be classified by their reaction with 2,2'-dipyridyl disulfide. .
