DOCUMENT TYPE: Journal CA Section: 7 (Enzymes) The transfer of [35S]sulfate from [35S]PAPS, by PAPS:chondroitin sulfate sulfotransferase, to various chondroitin sulfates with different degrees of sulfation and mol. wts. is reported. Analyses by digestion with chondroitinase AC and specific 4- or 6-sulfatases indicate that the sulfation occurs only in position 6 of the nonsulfated N-acetylgalactosamine moiety. The 50-70% desulfated chondroitin 4/6-sulfates are 2-fold better sulfate acceptors than totally desulfated chondroitin, and the affinity of the sulfotransferase increases markedly from the octa- to the decasaccharide. These results suggest that sulfation increases sharply only after the growing polysaccharide contains ~10 sugar residues, in the early stages of polymn., and that the sulfation of chondroitin sulfate may be a process in which the addn. of some sulfate groups facilitates further sulfation. .

Antibodies for chondroitin (I) and chondroitin 4-sulfate (II) were specific but the one for chondroitin 6-sulfate (III) cross-reacted with I. Indirect immunofluorescence staining with these antibodies required pretreatment with glycosaminoglycan-degrading enzymes. The 3 forms of chondroitin recognized by these antibodies have distinct and after separable distribution. In rats I was virtually restricted to cartilage, which also contained II and III. 9007-27-6 and 24967-94-0 which are cas registry numbers of chemicals are mentioned. In aorta, the intima and media contained III, whereas the adventitia and media contained II. It appeared that the proteoglycans of other tissues may predominantly carry only 1 type of chondroitin sulfate chain. Dermatan sulfate was widely distributed in loose connective tissue, esp. in the dermis; it was differentiated from II by pretreatment of tissue sections with chondroitinase AC. .
