DOCUMENT TYPE: Journal CA Section: 1 (Pharmacodynamics) The intestinal absorption of quinidine (I) [56-54-2] in rats was inhibited markedly by Na dextran sulfate [9011-18-1]. Na carboxymethylcellulose [9004-32-4] and Na alginate [9005-38-3] showed weaker inhibitory effects. The inhibitory effect of dextran sulfate was reflected in the blood level patterns obsd. after I was administered into the small intestine by the loop method; the time of the max. blood level was delayed, and after the max. was reached, the blood levels of I were higher than normal. A parallel relationship was established between the inhibitory effect of the polysaccharides on the intestinal absorption of I and the binding ratios of I-polysaccharide systems. In the physiol. pH region, dissocn. of the quinuclidine ring of I appeared to play an important role in the ionic binding of I and dextran sulfate.

(525 R) increased the no. of endogenous colonies in the spleen owing to a mitogenic effect on proliferation of colony forming units. None of the prepns. affected the no. of exogenous colonies when injected together with syngeneic bone marrow from intact donors. Dextran sulfate increased by 2.7-fold the no. of endogenous colonies in the spleens of unequally irradiated mice, mostly due to migration of colony-forming units from protected bone marrow sites. Poly A:U and yeast RNA were ineffective in this type of expt. The ability of dextran sulfate to increase migrational potencies of hemopoietic stem cells may be essential in the mechanism of its adjuvant activity.
