An example of the soaking soln. contg. invertase, mutarotase, glucose oxidase, peroxidase, N,N-dimethylaniline, 4-aminoantipyrine, albumin, and poly(vinyl alc.) in 0.1M phosphate buffer (pH 5.6) is given. The test paper strip contg. the above components detect sucrose at a detection limit of 0.01% and showed excellent coloration for sucrose in the concn. range of 0.01-4%. .

The 1st electrode is for H2O2 detection in sucrose detn. and consists of a thin Pt film-coated porous polycarbonate membrane in which glucose oxidase, mutarotase, and b-fructosidase are immobilized. The 2nd electrode is for eliminating interfering components, e.g., a- or b-glucose, and consists of a thin Pt film-coated porous membrane with immobilized glucose oxidase and mutarotase. These 2 electrodes are layered and attached to a plastic cylinder which contains buffer and ref. electrode. The 2nd electrode faces the sample soln. Thus, when a- or b-glucose is present in a sample, it is hydrolyzed by enzymes in the 2nd electrode and eliminated, and only sucrose reaches the 1st electrode at which H2O2 is produced after a series of enzyme reactions and detected by the Pt electrode. When ascorbic acid is present in a sample, it is oxidized at the 2nd Pt electrode. The current increase and the concn. of sucrose are linearly related. .
