DOCUMENT TYPE: Journal CA Section: 12 (Nonmammalian Biochemistry) Exts. of cerebral and pleuropedal ganglia from 2 terrestrial slugs, Ariolimax columbianus and Limax maximus, and from the marine opisthobranch, Aplysia californica, contain immunoreactivity resembling that of a vasotocin (I) or vasopressin (II). RIAs using several antisera indicate that the immunoreactivity is not due to I, II, or any other known naturally occurring neurohypophyseal peptide. Immunoreactivity of exts. on a relatively nonspecific II antiserum is well correlated with activity on antidiuretic assays on rats. Both immunoreactivity and antidiuretic activity are adsorbed onto bovine neurophysin affinity columns. Thus, these exts. contain 31 peptides that closely resemble the vertebrate antidiuretic hormones, I and II, both immunol. and pharmacol. The amts. of immunoreactivity and antidiuretic activity in ganglion exts. do not appear to change during dehydration and rehydration. Although both ganglionic exts. and I stimulate exudation of fluid across the slug body wall, the present expts. provide no evidence that the I-like material(s) in these ganglia may participate as neurotransmitters or hormones in the regulation of fluid balance. This remains an attractive hypothesis. .

(0.025 NL equiv./mL) stimulated a 425% increase in the release of glucagon [9007-92-5] from islets isolated from the pancreas of the rat. Gel filtration of the ext. produced an elution profile of glucagon-releasing activity that was superimposable on the profiles of oxytocin (OT) [50-56-6] and AVP [113-79-1]. Synthetic OT and AVP each elicited a concn.-dependent stimulation of glucagon release but failed to influence insulin [9004-10-8] release in medium 199 contg. 5.6 mM glucose. They were effective at 0.2 ng/mL (+55, +50%) and produced a striking increase (5-7-fold) at 20 ng/mL. The response to each peptide was greatly diminished in the presence of a higher concn. of glucose (11 mM). Lysine vasopressin [50-57-7], desamino-D-arginine vasopressin [16679-58-6], [1,6-aminosuberyl, 8-arginine] vasopressin [34786-77-1], vasotocin [9034-50-8], and AVP are equipotent peptides, whereas desamino-D-arginine vasopressin [16679-58-6], pressinoic acid [35748-51-7], and angiotensin II [11128-99-7] were inactive. Injections of AVP (1 mg, i.v.) produced a rapid increase in peripheral glucagon (185% in 5 min). The response to OT injection was less rapid (105% in 15 min), but in each case elevation of insulin was also obsd. Thus, OT and AVP can act directly on the endocrine pancreas thereby explaining some of the metabolic actions of these peptides. .
