Hepatoma cells converted 5-fluoro [14C]uridine to 5-fluorouridine diphospho glucose (FUDP glucose) [79965-09-6] FUDP-galactose [92748-40-8], FUDP-N-acetylglucosamine [87810-49-9], FUDP-N-acetylgalactosamine [92934-61-7], and trace amts. of FUDP-glucuronate [92748-41-9], as analyzed by different systems of high-performance liq. chromatog. 5-Fluoro[14C]uridine and 14C-labeled uridine [58-96-8], at concns. of 5 mM in the culture medium, were phosphorylated by the cells during 60 min to similar amts. of 5-fluorouridine 3'-triphosphate [92748-37-3] and UTP [63-39-8], resp., while the synthesis of [14]FUDP-sugars was reduced to 14% as compared to that of [14C]UDP-sugars. FUDP-sugars, synthesized by chem. and enzymic procedures, were assayed in vitro as substrates for enzymes of UDP-sugar metab. Km And V values in a range comparable to that of the resp. UDP-sugars were detd. for FUDP-sugars in the reactions catalyzed by UDP-glucose pyrophosphorylase [9026-22-6], galactose-1-phosphate uridylyltransferase [9016-11-9], UDP-glucose 4-epimerase [9032-89-7], UDP-N-acetylglucosamine 2-epimerase [9037-71-2], glycogen synthase [9014-56-6], and UDP-glucose dehydrogenase [9028-26-6]. Expts. in hepatoma cells and with enzymes in vitro have revealed addnl. reactions of FUDP-sugar metab. demonstrating a metabolite pattern analogous to that of UDP-sugars. The amts. of FUDP-sugars formed relative to UDP-sugars in intact cells were smaller than suggested on the basis of their kinetic comparison in vitro. .
