DOCUMENT TYPE: Journal CA Section: 64 (Pharmaceutical Analysis) To detn. folic acid (I) [59-30-3] in Polfamix L, a 1-g sample contg. 150 g I was dissolved in 10 mL 0.05M NaOH and 15 mL 0.05M HCl, and the soln. purified on a column packed with Florisil or Fuller's earth. I was eluted with boiling 4% Na2B4O7 and oxidized at pH 3 with 4% KMnO4 to 2-amino-4-hydroxypteridine-6-carboxylic acid [948-60-7], whose fluorescence was excited at pH 4-4.5 at 365 nm and measured at 470 nm.

(HPLC) assays for folic acid (I) [59-30-3] were developed. In the thin-layer procedure, the I band was extd. from the silica gel after development and detd. spectrophotometrically by an oxidn. and Bratton-Marshall sequence. A column packed with octadecylsilane chem. bonded to microparticulate silica gel was used for the HPLC assay. Potentiometric detn. of water in I samples was necessary to obtain accurate purity values. In a comparison of 4 assay methods, a direct colorimetric method gave precise, but unspecific, results, while the USP XIX colorimetric method lacked both precision and specificity. The 2 chromatog. methods were both precise and specific and gave the same assay results. Both methods sepd. p-aminobenzoic acid [150-13-0], N-(p-aminobenzoyl)-L-glutamic acid [4271-30-1], 2-amino-1,4-dihydro-4-oxo-6-pteridinecarboxylic acid [948-60-7], 2-amino-4(1H)-pteridinone [2236-60-4], and several unidentified impurities from I.
