Activation of mutagenic and carcinogenic heterocyclic amines by S-9 from the liver of a rhesus monkey
Activation of mutagenic and carcinogenic heterocyclic amines by S-9 from the liver of a rhesus monkey. Ishida, Y.; Negishi, C.; Umemoto, A.; Fujita, Y.; Sato, S.; Sugimura, T.; Thorgeirsson, S. S.; Adamson, R. H. (Natl. Cancer Cent. Res. Inst., Tokyo 104, Japan). Toxicol. In Vitro, 1(1), 45-8 (English) 1987. CODEN: TIVIEQ. ISSN: 0887-2333. DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Section cross-reference(s): 17 The mutagenicity of a series of heterocyclic amines isolated from various cooked foods and from pyrolyzates of amino acids and protein was assayed in Salmonella typhimurium TA98 in the presence of an S-9 fraction prepd. 77094-11-2 and 26148-68-5 are also in the experiment. from the liver of an untreated male rhesus monkey (Macaca mulatta). All the compds. showed mutagenicity, which ranged from 1 to 2500 revertants/mg chem./mg S-9 protein. 2-Amino-3,4-dimethylimidazo[4,5-f]quinoline (I) [77094-11-2] showed the highest specific mutagenic activity, followed by 2-amino-3,7,8-trimethylimidazo[4,5-f]quinoxaline [92180-79-5], 2-amino-3-methylimidazo[4,5-f]quinoline [76180-96-6], 2-amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline [95896-78-9], 3-amino-1-methyl-5H-pyrido[4,3-b]indole [62450-07-1], 3-amino-1,4-dimethyl-5H-pyrido[4,3b]indole [62450-06-0], 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline [77500-04-0], 2-amino-6-methyldipyrido[1,2-a:3',2'-d]imidazole [67730-11-4], and 2-amino-3-methyl-9H-pyrido[2,3-b]indole [68006-83-7]. 2-Amino-9H-pyrido[2,3-b]indole [26148-68-5] and 2-aminodipyrido[1,2-a:3',2'-d]imidazole [67730-10-3] also showed definite but weak mutagenicity. These specific mutagenic activities were very close to those of some heterocyclic amines assayed with the S-9 of human liver, slightly lower than those detd. with the S-9 of untreated rat liver, and much lower than those detd. with S-9 from the liver of the untreated hamster or mouse. The possible carcinogenicity of these heterocyclic amines in primates including humans was discussed. .
Analysis of mutagenic imidazo[4,5-f]quinolines and -quinoxalines (IQ compounds)
Analysis of mutagenic imidazo[4,5-f]quinolines and -quinoxalines (IQ compounds). Comparison of electrochemical and ultraviolet detection. Grivas, Spiros; Nyhammar, Tomas (Dep. Chem. Mol. Biol., Swed. Univ. Agric. Sci., Uppsala S-75007, Swed.). Mutat. Res., 142(1-2), 5-8 (English) 1985. CODEN: MUREAV. ISSN: 0027-5107. DOCUMENT TYPE: Journal CA Section: 17 (Food and Feed Chemistry) 3-Methyl-3H-imidazo[4,5-f]quinolin-2-amine [76180-96-6] 3,4-dimethyl-3H-imidazo[4,5-f]quinolin-2-amine [77094-11-2] 3,8-dimethyl-3H-imidazo[4,5-f]quinoxalin-2-amine [77500-04-0] and 3,4,8-trimethyl-3H-imidazo[4,5-f]quinoxalin-2-amine [95896-78-9], all known potent mutagens, have been sepd. by reversed-phase HPLC on a 250 ′ 4.6 mm i.d. column filled with Nucleosil C18, 5-m particle size, with MeOH-0.10M phosphate buffer, pH 7.3 () as mobile phase and detected by UV detection and electrochem. detection. The limits of detection were 2.5 pmoles for UV detection and 0.5-1.5 pmoles for electrochem. detection. The method is suitable for use in detecting these mutagens in beef exts. and other foods and food models.
