DOCUMENT TYPE: Conference CA Section: 2 (Mammalian Hormones) The effects of gestrinone [16320-04-0] and danazol [17230-88-5] on the LH [9002-67-9] response to LH-RH [9034-40-6] in rat anterior pituitary cells in primary culture were compared with those of the potent progestogen promegestone [34184-77-5], the androgen 5a-dihydrotestosterone [521-18-6], and the androgen with some progestational activity metribolone [965-93-5]. In vitro, on LH release, gestrinone was as effective as potent progestogens in stimulating LH release in estrogen-primed cultures and inhibiting it in unprimed cultures. In vivo, gestrinone inhibited LH secretion in ovariectomized rats to an extent similar to potent androgens. It inhibited both the basal plasma LH level and the pituitary sensitivity to LH-RH. Thus, gestrinone exerted progestational and androgenic activities at the anterior pituitary level on the control of LH-RH-induced LH secretion in female rats. Danazol was much less effective.

When 5a-dihydrotestosterone (DHT) [521-18-6] was the ligand, a curvilinear Scatchard plot was seen, which was resolved into 2 components: (1) the androgen receptor (AR), Kd = 0.12-0.44 nM; and (2) a low affinity species, Kd = 6.3-28 nM. The same cytosol demonstrated only the 1st type of binding for 3H-labeled methyltrienolone (MTr) [965-93-5], Kd = 0.10-0.40 nM. The AR (i.e., [3H]MTr binding activity) eluted at 440,000 daltons (d) by gel filtration chromatog. in prelabeling and postlabeling expts. When the ligand was [3H]DHT, binding activity in the 10,000-45,000 d range was seen in addn. to AR. Thus, saturable nonreceptor steroid binding was seen for DHT but not for MTr. The latter is the preferred ligand for the study of the AR in this system.
