DOCUMENT TYPE: Journal CA Section: 1 (Pharmacology) The pharmacokinetics of diphenidol (I) [972-02-1] (100 mg/kg, via gastric infusion) were studied in rats. I was rapidly absorbed and appeared in the blood and various tissues; the Ka and Kee (absorption and elimination consts., resp.) values for I were 1.24 and 0.124 h-1, resp., and the blood I was cleared in a 2-compartmental fashion with t1/2a and t1/2b values of 3.16 and 9.16 h, resp. The absorbed I was found mainly in the heart and liver, followed by spleen, lung, kidney, muscle, brain, and adipose tissues; but the tissue I levels were always lower than blood levels. Only 2.8% of the administered I was found in the urine 36 h after gastric infusion. No unmetabolized I was detectable in the urine 48 h after administration; instead, 3 metabolites of I were detected in the urine.
