DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) After i.p. injection of p-nitrophenylarsonic acid (I) [98-72-6] (30 mg/kg) into rats, N-acetyl-.beta.-glucosaminidase (II) [9012-33-3], .beta.-galactosidase (III) [9031-11-2], and .beta.-glucosidase (IV) [9001-22-3] activities in kidney homogenates increased; the activity of II increased by 30% within 24 h of I injection, remained high for a further day and then decreased as did the activities of III and IV. Within 48 h of I injection, the urinary isoenzyme patterns of II, III, and IV were identical to those found in the kidney. A decrease in enzyme latency within the first 24 h was the earliest change in the properties of the kidney subcellular fractions isolated after I injection. The total activity recovered in all the subcellular fractions was increased in the initial 24 h of the expt. but then fell rapidly, particularly in the lysosomal and sol. fractions. Thus, the initial stimulation of lysosomal enzyme activity was accompanied by a marked decrease in latency which may be a prelude to a loss of hydrolytic enzymes into the cytosol and then into the urine. IV activity found in the urine in tubular necrosis originates from the kidneys.
