DOCUMENT TYPE: Journal CA Section: 4 (Toxicology) Nitrobenzene [98-95-3] increases metHb formation when incubated with native Hb but not when incubated with red blood cell suspensions. These expts. were designed to det. if transport of nitrobenzene across the red blood cell membrane is a limiting factor for metHb prodn. by red blood cell suspensions. Incubation of 14C-labeled-m- [99-65-0], o- [528-29-0] or p-dinitrobenzene [100-25-4] but not mononitrobenzene, with red blood cell suspensions caused a time-dependent increase in metHb. All 3 dinitrobenzenes and mononitrobenzene crossed the red blood cell membrane and accumulated in the erythrocytes after only 1 min of incubation. Incubation of mononitrobenzene with hemolyzates did not result in metHb prodn. Incubation of red blood cells with the dinitrobenzenes or mononitrobenzene for 1 min and 10 min at 4° did not influence red blood cell uptake of the nitrobenzenes, suggesting that these compds. do not enter the red blood cell by an active process. Dinitrobenzene-induced metHb prodn. was markedly inhibited at 4°, and may be a result of decreased interaction with Hb and(or) decreased metab. to reactive intermediates which mediate metHb prodn. Thus, red blood cell transport of nitrobenzene is not the limiting factor in metHb prodn. in vitro.

The best results were obtained with PhNO2 in alk. medium as the oxidizing agent at 160°. With this reagent, 50-90% (based on coal wt.) water-sol. products were obtained. They provided insight into the origin and structure of coal.
