General Description

3,4-dihydroxybutanoic acid, also known as tartronic acid, is a four-carbon organic compound with two hydroxyl groups and a carboxylic acid group. It is a colorless crystalline solid with a slightly sour taste and is often found in the form of its salt, potassium tartronate. 3,4-dihydroxybutanoic acid is a key intermediate in the Krebs cycle, where it plays a crucial role in the oxidation of glucose and the production of energy in the form of adenosine triphosphate (ATP). It is also used in the synthesis of various pharmaceuticals and can be found in certain fermented food products. Additionally, it has potential applications in the production of biodegradable polymers and as a chelating agent in metal ion sequestration.

According to the present invention, by using 4-halogeno-3-hydroxybutanamide as a substrate in quaternary amination reaction with trialkylamine which is an important step in betaine (such as carnitine) preparation processes, it becomes possible to reduce the production of crotonic acid derivatives (the major by-product) greatly compared to conventional processes. Consequently, it becomes possible to prepare a betaine, such as carnitine, at a high yield. The present invention also relates to a process for preparing a betaine represented by formula (1) below, comprising a step of quaternary aminating an amide represented by formula (2) below: wherein A1, A2 and A3 individually represent a C1-C20 hydrocarbon group which may have a substituent(s); and X1 is a halogen atom.

The present invention presents a method for preparation of S-3,4-dihydroxy butyronitrile expressed by the structural formula (II) STR1 characterized by causing R-3-chloro-1,2-propanediol expressed by the structural formula (I) STR2 to react with a cyanating agent. According to this invention, optically active 3,4-dihydroxy butyronitrile and 3,4-dihydroxy butyric acid derivatives may be manufactured economically and efficiently.
