A series of 2-amino substituted benzoxazinones were prepared that potently inhibit human cytomegalovirus protease in vitro. Selected compounds demonstrate antiviral activity in cell culture, selectivity relative to chymotrypsin and elastase and stability with respect to hydrolysis in human plasma.
