2-chlorobenzimidazoles in which the nitrogen is substituted by R can react in an atypical way. When , heating with NaOH in DMF results in unexpected hydration of the triple bond. When R=CH2-CO-CH3, nucleophilic reagents (CH3O-, CN-, H-) attack at first the carbonyl function, forming an oxanion which cyclises by substitution of Cl.Compounds with these cyclic structures are sometimes isolable or can react further.Mechanisms are proposed for these reactions.
