The invention discloses a preparation method of tryptophan oxide as shown in a formula I which is described in the specification. According to the preparation method, N-tert-butyloxycarbonyl-L-tryptophan is used as a starting raw material, and is subjected to esterification, oxidation and deprotection so as to obtain the tryptophan oxide . According to the invention, the whole process route is reasonable in design, raw materials are low in cost, reaction conditions are mild, and operability is high; and the prepared tryptophan oxide can be used for quality control of commodities containing tryptophan components and has important application value.
