Glufosfamide and bromoglufosfamide are synthesized by reacting the corresponding tetracetyl derivatives with MeOH and a catalytic amount of MeO(-). Tetraacetyl glufosfamide is obtained by reacting a tetraacetyl trichloroacetamidate intermediate of glucose, ifosfamide mustard, and an acid in an anhydrous polar solvent.
