The invention provides a synthesis method of 5-hydroxymethyl tetrazole and derivatives thereof. The preparation method comprises the following steps: taking Trt protected carboxylic ester or aldehydecompounds as raw materials to react to obtain 5-hydroxymethyl tetrazole derivatives; the whole synthesis route is good in step repeatability, mild in operation condition, high in safety and beneficialto scale-up production and industrial popularization; after-treatment does not generate toxic waste liquid containing cyanide ions and has no pollution to the environment so that the production safety level and the production cost are reduced, application of green and environment-friendly industrial production is facilitated, and wide application prospects are achieved.

Cheap and safe: Conventional methods to prepare tetrazoles employ dangerous, toxic reagents. A new route to these heterocycles (see scheme) uses inexpensive and nontoxic dialkyl aluminum azides. The cycloaddition occurs under mild conditions and tolerates a variety of functional groups. The low cost and ecocompatibility make this process attractive for large-scale preparation.
