Protein Modification at Tyrosine with Iminoxyl Radicals
Post-translational modifications (PTMs) of proteins are a biological mechanism for reversibly controlling protein function. Synthetic protein modifications (SPMs) at specific canonical amino acids can mimic PTMs. However, reversible SPMs at hydrophobic amino acid residues in proteins are especially limited. Here, we report a tyrosine (Tyr)-selective SPM utilizing persistent iminoxyl radicals, which are readily generated from sterically hindered oximes via single-electron oxidation. The reactivity of iminoxyl radicals with Tyr was dependent on the steric and electronic demands of oximes; isopropyl methyl piperidinium oxime 1f formed stable adducts, whereas the reaction of tert-butyl methyl piperidinium oxime 1o was reversible. The difference in reversibility between 1f and 1o, differentiated only by one methyl group, is due to the stability of iminoxyl radicals, which is partly dictated by the bond dissociation energy of oxime O-H groups. The Tyr-selective modifications with 1f and 1o proceeded under physiologically relevant, mild conditions. Specifically, the stable Tyr-modification with 1f introduced functional small molecules, including an azobenzene photoswitch, to proteins. Moreover, masking critical Tyr residues by SPM with 1o, and subsequent deconjugation triggered by the treatment with a thiol, enabled on-demand control of protein functions. We applied this reversible Tyr modification with 1o to alter an enzymatic activity and the binding affinity of a monoclonal antibody with an antigen upon modification/deconjugation. The on-demand ON/OFF switch of protein functions through Tyr-selective and reversible covalent-bond formation will provide unique opportunities in biological research and therapeutics.

General Asymmetric Synthesis of Densely Functionalized Pyrrolidines via Endo-Selective [3+2] Cycloaddition of β-Quaternary-Substituted Nitroalkenes and Azomethine Ylides
A scalable endo-selective synthesis of 2,3,4,5-tetrasubstituted pyrrolidines via cycloaddition of nitroalkenes and azomethine ylides is reported using a P,N-type ferrocenyl ligand and [Cu(OTf)]2·C6H6. The robust method is tolerant of a wide range of functionalities, including rarely reported quaternary nitroalkene substitution and heteroaromatic and hindered ortho-substituted arenes on the azomethine ylide. Subsequent transformations highlight the utility of the method in the synthesis of densely functionalized small molecules suitable for fragment-based drug discovery and the cystic fibrosis C2-corrector clinical candidate ABBV-3221.

Method for preparing 2-amino-2-methyl-1-propanol
The invention relates to a method for preparing 2-amino-2-methyl-1-propanol. The method comprises the following steps: 1) enabling 2-chloropropane and sodium cyanide to react to generate 2-methyl propionitrile; 2) performing aldol condensation reaction on the 2-methyl propionitrile and formaldehyde so as to generate 2,2-dimethyl-3-hydroxy propionitrile; 3) performing hydrolysis reaction on 2,2-dimethyl-3-hydroxy propionitrile so as to generate 2,2-dimethyl-3-hydroxy propanamide; and 4) performing Hofmann degradation on the 2,2-dimethyl-3-hydroxy propanamide, thereby obtaining 2-amino-2-methyl-1-propanol. The method is cheap and easy in obtaining of raw materials of different steps, simple in reaction process, free of hash reaction condition, remarkable in cost advantage, high in yield, low in pollution and easy in product purification.

The 2-cyano-2,2-dimethylethanimine-N-oxymethyl group for the 2′-hydroxyl protection of ribonucleosides in the solid-phase synthesis of RNA sequences
The reaction of 2-cyano-2-methyl propanal with 2′-O- aminooxymethylribonucleosides leads to stable and yet reversible 2′-O-(2-cyano-2,2-dimethylethanimine-N-oxymethyl)ribonucleosides. Following N-protection of the nucleobases, 5′-dimethoxytritylation and 3′-phosphitylation, the resulting 2′-protected ribonucleoside phosphoramidite monomers are employed in the solid-phase synthesis of three chimeric RNA sequences, each differing in their ratios of purine/pyrimidine. When the activation of phosphoramidite monomers is performed in the presence of 5-benzylthio-1H-tetrazole, coupling efficiencies averaging 99 % are obtained within 180 s. Upon completion of the RNA-chain assemblies, removal of the nucleobase and phosphate protecting groups and release of the sequences from the solid support are carried out under standard basic conditions, whereas the cleavage of 2′-O-(2-cyano-2,2-dimethylethanimine-N-oxymethyl) protective groups is effected (without releasing RNA alkylating side-products) by treatment with tetra-n-butylammonium fluoride (0.5 m) in dry DMSO over a period of 24-48 h at 55 °C. Characterization of the fully deprotected RNA sequences by polyacrylamide gel electrophoresis (PAGE), enzymatic hydrolysis, and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry confirmed the identity and quality of these sequences. Thus, the use of 2′-O-aminooxymethylribonucleosides in the design of new 2′-hydroxyl protecting groups is a powerful approach to the development of a straightforward, efficient, and cost-effective method for the chemical synthesis of high-quality RNA sequences in the framework of RNA interference applications. A NOVEL SYNTHESIS OF 3,3-(SPIRO)SUBSTITUTED AZETIDINES
A smooth and efficient new synthesis for 3,3-disubstituted azetidines, starting from readily available nitriles, was estabilished: α-hydroxymethylation of the starting materials, followed by O-tosylation and LiAlH4-reduction of the key intermediates thus obtained, led - via spontaneous cyclization of the intermediate amino derivatives - to 3,3-disubstituted azetidines.Scope and limitations of this new method were studied with respect to generalized applicability: the target compounds were accessible in good yields for a variety of starting materials (cyclic and acyclic di(hetero)aryl, (hetero)arylalkyl, dialkyl, as well as basic moieties).The products thus obtained may be of interest for ensuing conversions due to their unblocked nitrogen.
