2-(1H-lndole-3-carbonyl)-thiazole-4-carboxamide derivatives and the corresponding imidazole, oxazole and thiophene derivatives and related compounds as aryl hydrocarbon receptor (AHR) agonists for the treatment of angiogenesis implicated disorders, such as e.g. retinopathy, psoriasis, rheumatoid arthritis, obesity and cancer, or inflammatory disorders. Disclosed is an aryl hydrocarbon receptor modulator of formula (I), and a pharmaceutically acceptable salt thereof, wherein R′ is H, CN, CH2(OH)R0, CmH2m+1, CnH2n-1, CnH2n-3, two Ra are independently H or two Ra together form ═O or ═N—W3—R1; A is a C6 to C10 aromatic ring unsubstituted or substituted with 1 to 3 R, or a C2-C10 heteroaromatic ring interrupted by 1 to 5 heteroatoms selected from N, O, and S or a 4 to 7 membered nonaromatic heterocyclic ring containing C═N and interrupted by 1 to 3 heteroatoms selected from N, O, and S, with either one unsubstituted or substituted with 1 to 3 R; Q is R, or is a C6 to C10 aromatic ring or a C2 to C10 heteroaromatic ring unsubstituted or substituted with 1 to 3 R and interrupted by 1 to 5 heteroatoms selected from N, O, and S; and R is Rc which is C-attached or RN which is N-attached. The compounds of formula (I) of the present invention can regulate AhR activity, and can be used to inhibit the growth of cancer cells and inhibit the metastasis and invasion of tumor cells.

- Alpha-ethyltryptamines as dual dopamine-serotonin releasers

The dopamine (DA), serotonin (5-HT), and norepinephrine (NE) transporter releasing activity and serotonin-2A (5-HT2A) receptor agonist activity of a series of substituted tryptamines are reported. Three compounds, 7b, (+)-7d and 7f, were found to be potent dual DA/5-HT releasers and were >10-fold less potent as NE releasers. Additionally, these compounds had different activity profiles at the 5-HT2Areceptor. The unique combination of dual DA/5-HT releasing activity and 5-HT2Areceptor activity suggests that these compounds could represent a new class of neurotransmitter releasers with therapeutic potential.

(matrix presented) Various tryptamines are captured by a vinylsulfonylmethyl polystyrene resin, generating a safety-catch linkage. β-Carbolines can be formed from 4 by a Pictet-Spengler reaction with the introduction of R1. Tryptamine 4 can also be derivatized by acylation or copper-mediated coupling to introduce R2. If X = Br, Suzuki coupling can be used to introduce R3. After derivatization, the indole derivatives are activated with methyl iodide and released under mild basic condition.

- Electron Impact and Chemical Ionization Fragmentation of 5-Methoxytryptamine and Some 6-Methoxy-β-carbolines

Electron impact (EI) and chemical ionization (CI) mass spectra of 5-methoxytryptamine (5-MT) and seven 6-methoxy-β-carbolines have been recorded and interpreted.Proposed fragmentation pathways are depicted on the basis of spectra of deuterium labelled analogs of 5-MT, 6-methoxy-1,2,3,4-tetrahydro-β-carboline (6-MeO-THBC) and 6-Methoxy-1-methyl-1,2,3,4-tetrahydro-β-carboline (6-MeO-1-Me-THBC).In the EI mass spectrum of 5-MT, the base peak m/z 160 is formed by the expulsion of CH4N.The positive ion so formed is suggested to have a quinolinium structure.In chemical ionization with CH4 as the reaction gas, the most abundant ion is produced by cleavage of NH3.The ion m/z 173 yields the base peak in the EI mass spectrum of 6-MeO-THBC.This ion is formed by a retro Diels-Alder reaction, an important fragmentation pathway of 6-MeO-1-Me-THBC, too. 6-MeO-harmalan easily loses one hydrogen atom in EI fragmentation followed by methyl loss from the methoxy group, this latter process yielding the base peak, m/z 197 in the EI mass spectrum of 6-MeO-harman.M+1 is the base peak in the CI mass spectra of all 6-MeO-β-carbolines.

The mass spectra of the four tryptamine derivatives, N-acetyl-5-methoxytryptamine (melatonin), N-acetyl-5-hydroxytryptamine (N-acetyl-serotonin), N,N-dimethyl-5-hydroxtryptamine (bufotenine) and N,N-dimethyl-5-methoxytryptamine (O-methylbufotenine), with specifically labeled [D4] aminoethyl sidechains have been measured. Comparison of these spectra with those of the unlabeled compounds enable the major fragmentations of the compounds to be defined.
