- Photoinduced viscosity control of lecithin-based reverse wormlike micellar systems using azobenzene derivatives

This report describes the controlled viscosity changes of photoresponsive reverse wormlike micellar systems formed by soybean lecithin (SoyPC), d-ribose, and azobenzene derivatives in decane. UV light irradiation produces a significant (150-fold) decrease in solution viscosity by triggering a structural transformation of the wormlike micelles. Subsequent visible light irradiation leads to recovery of the initial micellar structure and elevated solution viscoelasticity. This dramatic, reversible variation in solution viscosity by light irradiation can be applied to cosmetics, personal care products, and device components.

A photoreversible switch based on a photoresponsive host-guest system consisting of dimethylamino calix[4]arene L and 4-(phenylazo)benzoic acid (O) is reported. The host L exhibited selective binding and release of O on UV and visible irradiation at 450 and 365 nm, respectively. Moreover, the photoresponsive host-guest complex was applied as a photocontrolled wettability switch on a functional micro/nanostructured silicon surface, and is thus promising for applications in sensors and microfluidic devices. Photoreversible switching of the host-guest system of dimethylamino calix[4]arene (L) and 4-(phenylazo)benzoic acid (O) is reported. Host L exhibited selective binding and release of O under UV and visible irradiation at 450 and 365 nm, respectively. Moreover, the L?O complex was applied as a photocontrolled wettability switch on a functional micro/nanostructured silicon surface with reversible switching between hydrophilic and hydrophobic states (see figure).

- Photosensitive and Photoswitchable TRPA1 Agonists Optically Control Pain through Channel Desensitization

Transient receptor potential ankyrin 1 (TRPA1) channel, as a nonselective ligand-gated cation channel robustly in dorsal root ganglion sensory neurons, is implicated in sensing noxious stimuli and nociceptive signaling. However, small-molecule tools targeting TRPA1 lack temporal and spatial resolution, limiting their use for validation of TRPA1 as a therapeutic target for pain. In our previous work, we found that 4,4′-(diazene-1,2-diyl)dianiline (AB1) is a photoswitchable TRPA1 agonist, but the poor water solubility and activity hinder its further development. Here, we report a series of specific and potent azobenzene-derived photoswitchable TRPA1 agonists (series 1 and 2) that enable optical control of the TRPA1 channel. Two representative compounds 1g and 2c can alleviate capsaicin-induced pain in the cheek model of mice through channel desensitization but not in TRPA1 knockout mice. Taken together, our findings demonstrate that photoswitchable TRPA1 agonists can be used as pharmacological tools for study of pain signaling.

- Light-controlled out-of-equilibrium assembly of cyclodextrins in an enzyme-mediated dynamic system

We show that the selective enzymatic synthesis of specific cyclodextrins can be modulated using light. We use enzyme-mediated dynamic combinatorial chemistry to generate a mixture of interconverting linear and cyclic α-1,4-glucans, and employ an azobenzene photoswitch as a template. Using UV or blue light to switch between photostationary states with different azobenzene cis/trans isomeric ratios, we can promote the out-of-equilibrium assembly of either α-cyclodextrin or β-cyclodextrin.
