The invention discloses a preparation method of glycin-1-14C, which comprises the following steps: (1) under the protection of inert gas, enabling anhydrous nitromethane and metal or a metal organic reagent to form a nitromethane salt solution in an anhydrous ether solvent, and then reacting the nitromethane salt solution with CO2 to obtain a crude product of nitroacetic acid-1-C, wherein the CO2 is obtained by acidolysis of barium carbonate C; and (2) under the action of a palladium-carbon catalyst, carrying out reduction reaction on the crude product of the nitroacetic acid-1-C and hydrogen, and after the reaction is finished, carrying out post-treatment to obtain the glycin-1-C. The method has the advantages of short synthesis steps, mild and controllable reaction conditions, no need of high temperature, high pressure and other harsh conditions, no need of use of highly corrosive, highly toxic and other dangerous chemicals, easy trace synthesis, low cost and high C utilization rate, and the finally purified product has the advantages of high purity, good state and small specific activity reduction.
