2-Methoxyethyl 4-(3-hydroxyphenyl)-7-(2-methoxyphenyl)-2-methyl-5-oxo-1,4,5,6,7,8-hexahydroquinolin is a cell-permeable oxo-tetrahydroquinolinecarboxylate compound that functions as a downstream Hh (Hedgehog) pathway blocker. It effectively inhibits Gli-dependent transcription activity in various cell cultures, including Sufu+/+ NIH 3T3 (shh-LIGHT2) and Sufu-/fibroblast cultures, with an IC50 of 1.5 and 3 μM, respectively. Its inhibitory effect is similar to that of two other Hh downstream blockers, HPI-3 and GANT-61. However, HPI-1 is the only compound that remains effective against Sufu-/fibroblasts overexpressing Gli1 or Gli2, with an IC50 of 6 and 4 μM, respectively. Additionally, HPI-1 inhibits ligand-independent Hh pathway activation in SmoM2-expressing primary murine CGNPs (Cerebellar Granule Neuron Precursors) and NIH 3T3 (smoM2-LIGHT) cultures.
