In an investigation of 4-amino-3,4-dihydronaphthalen-1(2H)-ones as novel modulators of allergic and inflammatory phenomena, we have investigated a series of cyclic analogues. Tertiary amines of structural types 9, 10, 20 and 21 were synthesised and evaluated for mast cell stabilising activity. In vitro and in vivo studies showed that of these compounds, the cyclohexenylamino derivatives of tetralone and benzosuberone of series 20 and 21 exhibited interesting activity both in vitro and in vivo.

Direct photoamination of phenanthrene, 9-methoxyphenanthrene, anthracene, naphthalene, and several substituted naphthalenes with ammonia or primary amines in the presence of m-dicyanobenzene occurs to give aminated dihydroarenes in fairly good yields. m-Dimethoxybenzene and biphenyl are photoaminated in lower yields.A suggested mechanism for the photoamination involves the nucleophilic attack of ammonia and amines on aromatic cation radicals generated by photochemical electron transfer to m-dicyanobenzene.The present photoamination is applied to direct introduction of various functionalized primary amines containing the vinyl, cyano, hydroxy, acetylamino, and ethoxycarbonyl groups.

The above tetrahydro-4-oxo-1-naphthyl-urea and certain derivatives thereof are animal growth promoting agents. This invention further relates to methods of preparation of said 1-benzoyl (and 1-substituted benzoyl) 3-(1,2,3,4-tetrahydro-4-oxo-1-naphthyl)ureas.
