- An efficient synthetic approach towards new 5,5’-diaryl-2,2’-bipyridine-based fluorophores

An efficient approach has been developed for the synthesis of 5,5′-diaryl-2,2′-bipyridines via their 1,2,4-triazine analogues. The notable advantages of the present method are: The possibility of varying the aromatic substituents in the positions 5 and 5′ of bipyridine core and the possibility for obtaining 2,2′-bipyridines bearing a fused cyclopentene core to increase the solubility in organic solvents. These 5,5′-diaryl-2,2′-bipyridines exhibited an intense emission in a range of ca. 422–521?nm in acetonitrile solution; depending on the nature of the aromatic substituents and the presence of annulated cyclopentene fragments. Apart from that, the significant bathochromic shifts of the both absorption and emission maxima were observed in comparison with a number of previously described similar structures. In some cases the significant increasing of the fluorescence quantum yields took place.

FeSO4-mediated direct arylation of heteroarenes with arylboronic acids in the presence of K2S2O8 has been developed. A slow addition of an aqueous solution of an iron complex was crucial in the arylation. Scope and limitation of the heteroarenes and arylboronic acids are discussed. Furthermore, the direct arylation was applied to the formal total synthesis botryllazine B and sodium channel inhibitor.

- Inhibition of 1-deoxy-d-xylulose-5-phosphate reductoisomerase by lipophilic phosphonates: SAR, QSAR, and crystallographic studies

1-Deoxy-d-xylulose-5-phosphate reductoisomerase (DXR) is a novel target for developing new antibacterial (including antituberculosis) and antimalaria drugs. Forty-one lipophilic phosphonates, representing a new class of DXR inhibitors, were synthesized, among which 5-phenylpyridin-2-ylmethylphosphonic acid possesses the most activity against E. coli DXR (EcDXR) with a K i of 420 nM. Structure-activity relationships (SAR) are discussed, which can be rationalized using our EcDXR:inhibitor structures, and a predictive quantitative SAR (QSAR) model is also developed. Since inhibition studies of DXR from Mycobacterium tuberculosis (MtDXR) have not been performed well, 48 EcDXR inhibitors with a broad chemical diversity were found, however, to generally exhibit considerably reduced activity against MtDXR. The crystal structure of a MtDXR:inhibitor complex reveals the flexible loop containing the residues 198-208 has no strong interactions with the 3,4-dichlorophenyl group of the inhibitor, representing a structural basis for the reduced activity. Overall, these results provide implications in the future design and development of potent DXR inhibitors.
