The invention discloses a preparation method of 5-fluoro-1H-pyrrolo[2,3-b]pyridine-4-carbaldehyde, comprising the following steps: dissolving 5-fluoro-1H-pyrrolo[2,3-b]pyridine in a solvent, and adding triisopropylchlorosilane under alkaline conditions for reaction to obtain 5-fluoro-1-triisopropylsilylpyrrolo[2,3-b]pyridine; dissolving 5-fluoro-1-isopropylsilylpyrrolo[2,3-b]pyridine in a solvent,adding a formylating reagent under alkaline conditions for reaction, and after the reaction, adding a desiliconization reagent for in situ deprotection to obtain 5-fluoro-1H-pyrrolo[2,3-b]pyridine-4-carbaldehyde. The preparation method for the 5-fluoro-1H-pyrrolo[2,3-b]pyridine-4-carbaldehyde of the invention is a two-step method with advantages of short preparation route, easy-to-obtain raw materials, simple operation, remarkably reduced production cost and no generated heavy metal solid waste. The method is suitable for large-scale production of 5-fluoro-1H-pyrrolo[2,3-b]pyridine-4-carbaldehyde.
