- 2-Methyl-1,2,3,4-tetrahydro-10H-pyrido[4,3-b][1,4]benzothiazines with analgesic activity

The sodium borohydride reduction of 2-methyl-10-substituted-10H-pyrido[4,3-b][1,4]benzothiazinium iodide oxalates in ethanol at -15°C affords the 1,2,3,4-tetrahydro derivatives. On the other hand, reaction of 10 acetyl 10H pyrido[4,3 b][1,4]benzothiazine with methyl chloroformate and sodium borohydride in methanol at -78° C gave the 1,2-dihydro derivative. The 1,2,3,4-tetrahydro compounds exhibited significant analgesic activity comparable to that of acetylsalicylic acid and dextropropoxyphene.
