The invention relates to a preparation method of N alpha-fluorenylmethoxycarbonyl-N-Epsilon-7-methoxycoumarin-4-acetyl-lysine/ornithine and mainly aims to solve the problems that a current method is complicated in treatment, lower in yield and not environment-friendly. According to the technical scheme, the preparation method of N alpha-fluorenylmethoxycarbonyl-N-Epsilon-7-methoxycoumarin-4-acetyl-lysine/ornithine comprises steps as follows: N alpha-fluorenylmethoxycarbonyl-N Epsilon-7-methoxycoumarin-4-acetyl-lysine/ornithine tert-butyl ester is produced from N alpha-fluorenylmethoxycarbonyl-lysine/ornithine tert-butyl ester and 7-methoxycoumarin-4-acetic acid through condensation under catalysis of a condensing agent, tert-butyl ester is removed from N alpha-fluorenylmethoxycarbonyl-N Epsilon-7-methoxycoumarin-4-acetyl-lysine/ornithine tert-butyl ester by use of trifluoroacetic acid, and a product, namely, N alpha-fluorenylmethoxycarbonyl-N-Epsilon-7-methoxycoumarin-4-acetyl-lysine/ornithine is obtained. The product is mainly used as a fluorescent probe or a marker.
