w/v milk (1: 500 dilutions; Cayman), p-p38 was diluted in TBS-T
with 5% w/v BSA (1: 500 dilutions; Cell Signaling Technology),
and α-tubulin was diluted in TBS-T with 5% w/v milk (1:12 000
dilutions; Oncogene Science). PVDF membranes were extensively
washed and incubated with a horseradish peroxide-conjugated
secondary antibody (1: 2000 dilutions) for 1 h at room tempera-
ture. Immuno-reactive bands were visualized with an enhanced
chemiluminescence substrate kit (Amersham Pharmacia Biotech,
GE Healthcare UK Ltd, Bucks, UK) according to the manufacturer’s
protocol.

Superoxide Detection: The superoxide anion was fluorometri- [14] S. H. Wu, C. T. Tseng, Y. S. Lin, C. H. Lin, Y. Hung, C. Y. Mou,
cally estimated using a fluorescent probe, dihydroethidine (DHE),
which is oxidized to a fluorescent intercalator, ethidium, by cel-
lular oxidants, in particular, superoxide radicals. Therefore, to
measure the superoxide anion generation in our system dihydro-
ethidine (DHE; 5 μM) was used. The increase in DHE fluorescence

upon paraquat (superoxide anion generator) stimulation indicated [17] S. P. Hudson, R. F. Padera, R. Langer, D. S. Kohane, Biomaterials
an increase in superoxide anion levels. After various experimental
treatments, fluorescence images were obtained with a fluores-
cence microscope (IX-71, Olympus) and quantitative data were
analyzed with a FACS Calibur flow cytometer.
