The study focuses on bacterial fatty acid synthesis (FAS), a process crucial for bacterial cell membrane formation and a promising target for antibacterial drugs due to its differences between bacterial and mammalian systems. The researchers isolated Aquastatin C through a series of chemical processes, including ethyl acetate extraction, silica gel column chromatography, and HPLC purification. They determined its structure using NMR spectroscopy and other analytical techniques. The study found that Aquastatin C, unlike its precursor Aquastatin A, does not inhibit bacterial enoyl-ACP reductase enzymes (FabI, FabK, and InhA) or show antibacterial activity against Staphylococcus aureus, methicillin-resistant S. aureus, and Streptococcus pneumoniae. This suggests that the 2-hydroxy-6-methyl benzoic acid ring, absent in Aquastatin C, is essential for the inhibitory activity of Aquastatin A. The findings provide insights into the structure-activity relationship of these compounds, highlighting the importance of specific structural features for their antibacterial efficacy.
