The study developed a competitive indirect enzyme-linked immunosorbent assay (ciELISA) for the detection of the organophosphorus insecticide acephate. Five different haptens mimicking acephate were synthesized and conjugated with carrier proteins to raise polyclonal antibodies in rabbits. These antibodies were then used to detect acephate in various samples. The study evaluated the effects of different assay conditions on sensitivity and found an IC50 value of 25 ng/mL for acephate, with a detection range of 5-140 ng/mL and a lowest detection limit of 2 ng/mL. The assay showed low cross-reactivities with structurally related insecticides, indicating high specificity. The chemicals used in the study included acephate, various haptens, bovine serum albumin (BSA), keyhole limpet hemocyanin (KLH), and organic solvents for assay optimization. These chemicals served to mimic the target analyte, carry the haptens in the assay, and adjust assay conditions to optimize sensitivity and specificity for detecting acephate residues in environmental and agricultural samples.
