The research focuses on the development of a general strategy for synthesizing several members of the calyxin family of natural products, which have significant antiproliferative activity against certain cancer cell lines. The study explores the Suzuki coupling reactions between 2,4,6-trialkoxyphenylboronic acids and enol triflates, aiming to synthesize a lactone template that could serve as a precursor for various calyxin analogues. The researchers found that the Suzuki coupling reaction with enol triflates yielded excellent results, while enol tosylates failed to produce the desired product. The study successfully synthesized a lactone template through the coupling reaction, demonstrating a feasible approach for the synthesis of calyxin analogues. However, limitations were observed regarding the choice of protective groups on the trialkoxyphenyl substituent. The findings lay the groundwork for the total synthesis of calyxin analogues 1–3 using the lactone template.
