The conversion involves the NADP⁺-dependent removal of the 21α-hydrogen by geissoschizine dehydrogenase, followed by cyclization to form the intermediate 4,21-dehydrogeissoschizine and subsequent reduction to yield the final product. This enzyme exhibits high substrate specificity and low activity compared to other enzymes in the ajmalicine pathway. The stereochemistry of the reaction ensures the formation of the 19-epi configuration, distinguishing it from ajmalicine.
