This research explores the application of immobilized penicillinacylase in the synthesis of the dipeptide sweetener aspartame. The purpose is to find a more selective and efficient method to replace the formyl group in the traditional synthesis route with a nitrogen protecting group that can be removed enzymatically. The researchers used penicillinacylase immobilized on Eupergit C beads to catalyze the conversion of N-phenacetyl aspartame into aspartame and phenylacetic acid at pH 7.5. The results showed that the enzyme selectively cleaved the phenacetyl group, and the rate of hydrolysis depended on the substituents at the α-carboxyl group of aspartic acid and the absolute configuration of aspartic acid.
