The study evaluates the cytotoxicity of novel 6-(4-benzamido-/4-phthalimido)-3-cyanopyridine derivatives against prostate carcinoma PC3, human hepatocellular carcinoma HepG2, and breast adenocarcinoma MCF-7 cell lines using the MTT assay, revealing that compounds 6, 7, 8, 12, and 13 exhibit significant cytotoxic effects, being 1.5-3.3 times more potent than the reference standard 5-FU. These compounds also show high selectivity towards cancer cells over normal human cells (WI-38). Further evaluation of their PIM-1 kinase inhibitory activity indicates that compounds 12 and 13 have the highest potency, with inhibition percentages of 73.64% and 76.43%, respectively. Apoptosis studies reveal that these compounds enhance caspase-3 activation and increase the Bax/Bcl-2 ratio, promoting apoptosis. Molecular docking studies and in silico predictions confirm the potential of compound 13 as a promising PIM-1 inhibitor with favorable pharmacokinetic properties. The findings suggest that these cyanopyridine derivatives could serve as effective anticancer agents, particularly against prostate cancer.
