The research focuses on the yeast Malassezia furfur, which is known to convert tryptophan into several indole compounds, one of which is malassezin. The purpose of the study was to identify and characterize malassezin as a novel agonist of the arylhydrocarbon receptor (AHR) from cultures of M. furfur. The researchers synthesized malassezin and found it to induce cytochrome P450, acting as an AHR agonist with an EC50 of 1.57μM in rat hepatocytes. The conclusions drawn from the study suggest that malassezin, the first alkaloid described from cultures of a lipophilic Malassezia yeast, could be a novel prototype of AHR agonists or potentially converted intracellularly into a potent AHR agonist such as indolo[3,2-b]carbazole (7).
