The invention discloses a preparation method of a mitochondrial targeting fluorescein derivative fluorescent probe, and belongs to the technical field of biology. The method comprises the steps: making fluorescein and hydrazine hydrate react to obtain a fluorescein hydrazine compound; adding the fluorescein hydrazine compound into an amino acid absolute ethyl alcohol solution to react to obtain a fluorescein hydrazine derivative; reacting the fluorescein hydrazine derivative with a mitochondrial targeting ligand to obtain a fluorescein hydrazine derivative-mitochondrial targeting substance; and reacting the fluorescein hydrazine derivative-mitochondrial targeting substance with recognition molecules to obtain the targeting mitochondrial fluorescein derivative fluorescent probe. The targeting mitochondrial fluorescein derivative fluorescent probe obtained by the invention has high recognizability on thiophenol and strong anti-interference detection capability; the positioning effect on the mitochondria is good, and the Pearson coefficient obtained by detecting the mitochondria is 0.96; and the mitochondrial membrane potential is obviously reduced.

The method comprises: preparing an ultraviolet sensor solution according to the claim 1, adding a nickel ion solution to be tested into the sensor solution, metering the volume of the solution, carrying out standing, testing the absorbanceat wavelength of 380 nm to 420 nm and acquiring a Ni concentration according to a standard curve. The fluorescein derivative-based probe has good selectivity to nickel ions, is easy to use in sewage treatment application and has good use effects.
