Description
SM 5887, also known as Amrubicin, is a completely synthetic anthracycline derivative used for the treatment of small and small-cell lung cancers. It mediates its growth inhibitory action via topoisomerase II inhibition and is activated in vivo by the formation of its 13-OH metabolite, amrubicinol, via reaction with carbonyl reductase. Amrubicinol is 10-100 fold more cytotoxic than amrubicin and has shown antitumor activity against non-small-cell lung cancers and untreated extensive stage small-cell lung cancers in phase II clinical trials. The drug has a smaller distribution-volume, shorter half-life in mice, and less chronic cardiotoxicity compared to doxorubicin. It is generally well tolerated with major adverse events being anaemia, leucopenia, thrombocytopenia, and neutropenia. The brand name for SM 5887 is Calsed.
