The invention discloses a novel compound--didansyl histidine, and a preparation method and application thereof. The chemical formula of didansyl histidine is shown in a formula I which is described in the specification, and didansyl histidine can be used as a fluorescent probe for detection of iron ions. The invention further discloses a method for fluorescence recovery of didansyl histidine after recognition of iron ions. Didansyl histidine has good water-solubility, is easy to prepare an aqueous solution and fluorescently stable and can conveniently detect the concentration of iron ions in an aqueous solution. Didansyl histidine is a high-sensitivity iron ion fluorescent probe, has an iron ion detection limit of 2.0 to 10 mol/L and can recognize iron ions at a pH value in a range of 2.50 to 10.35, and common metal ions do not affect specific recognition of iron ions by didansyl histidine. As the iron ion fluorescent probe under an acidic condition, didansyl histidine has the characteristics of a wide application scope, good recognizability and god accuracy and is suitable for popularization and application.
