Synthesis, Neuroleptic and Antiinflammatory Activities of 4a,11a-cis- and trans-2--1,2,3,4,4a,5,11,11a-octahydro-6H-pyridocarbazoles and Related Derivatives

A number of 2-substituted derivatives (5a-15b) of 4a,11a-cis/trans-2H-1,2,3,4,4a,5,11,11a-octahydro-6H-pyridocarbazoles (3a,b and 4b), obtained in seven steps, have been synthesized and studied for their pharmacological activities.Both cis- and trans-2--6H-octahydro-pyridocarbazoles (7a and 8b) exhibit tranquilising activity comparable to chlorpromazine while trans-8b also shows potent antiinflammatory activity at the tranquilising dose.The study, however, suggests that N-5 in centbutindole is not an essential feature for tranquilising activity.
