9-(3-Substituted-2-hydroxypropyloxy)acridines (5-11) and 9-(2-hydroxy-3-morpholinopropyloxy)-1,2,3,4-tetrahydroacridine (17) have been synthesised via the epoxy intermediates 9-(2,3-epoxypropyloxy)acridine (4) and the corresponding tetrahydroacridine (15).A few 9-(thiocyanato, phenylcarbamoyloxy and substituted amino)-tetrahydroacridines (21, 23 and 24-27) and 2,3-epoxypropyl N-phenylanthranilate (3) have also been prepared.Compounds 15, 17, 22, 24 and 26 show in vitro antiamoebic activity at 16 - 62.5 μg/ml dose while the standard drug metronidazole is found antiamoebic at 4 μg/ml.Compounds 25-27 exhibit local anaesthetic activity at 0.2percent concentration.
