The invention belongs to the technical field of predrug carrier preparation and discloses a preparation method of a glucan-QT (quercetin) predrug polymer. According to the preparation method, acyl chloride groups are introduced through reactions including protection, selective substitution, deprotection, acylating chlorination and the like of hydroxyl of QT and are linked with glucan, and the glucan-QT predrug polymer is prepared. The preparation method is convenient and easy to implement and mild in reaction conditions. A basis is provided to improve the water solubility of an ideal drug carrier constructed to study a derivative of QT, and an earlier scheme is provided to improve the bioavailability.
