(2S)-[(2S)-[(2R)-AMINO-3-MERCAPTO]-PROPYLAMINO-(3S)-METHYL]PENTYLOXY-3-PHENYLPROPIONYL-METHIONINE SULFONE ISOPROPYL ESTER BIS HYDRCHLORIDE, also known as L-744,832, is a cell-permeable potent and selective thiol-containing peptidomimetic farnesyltransferase (FTase) inhibitor with anti-tumor properties. It rapidly blocks p70S6K activation and DNA synthesis and promotes apoptosis in transgenic mice. L-744,832 induces p21 expression and cell cycle arrest in the G1 phase. It displays synergistic effects with paclitaxel and epothilones in inhibiting tumor growth. Additionally, it mimics many of the effects of Rapamycin and may be effective against tumors that exhibit inappropriate activation of the mTOR/p70S6K pathway. L-744,832 is a white crystalline solid.

Used in Oncology:
L-744,832 is used as a ras farnesyltransferase inhibitor for inducing tumor regression in transgenic mice with multiple oncogenic mutations by mediating alterations in both cell cycle control and apoptosis. It induces p21 expression and arrests cells at G1, causing enhanced mitotic sensitivity to taxol.
Used in Drug Development:
L-744,832 is used as a research compound for studying the effects of farnesyltransferase inhibition on tumor growth and the potential synergistic effects with other chemotherapeutic agents such as paclitaxel and epothilones.
