Six analogues of leucine-/methionine-enkephalinamides have been synthesised by substitution of D-amino acids at position-2 and Nle or Eth at position-5 by solid phase technique employing mainly the base labile 9-fluorenyl-methyloxycarbonyl group for Nα-protection.One of the analogues, tyrosyl-D-norvalyl-glycyl-phenylalanyl-ethioninamide is found to be 21.6 times more potent than morphine in the guinea pig ileum assay.
